·本片是首部完全在纽约拍摄的《蜘蛛侠》电影，也是在纽约拍摄的最大预算的影片。
·本集的配乐由大师汉斯·季默与美国创作型说唱歌手法瑞尔·威廉姆斯联手打造。
·奥斯卡影帝杰米·福克斯在片中扮演的是电光人，奥斯卡最佳男配角克里斯·库珀扮演的是奥斯本，这是奥斯卡获奖演员第一次扮演蜘蛛侠的死对头。
·漫画中，格温被老绿魔（诺曼奥斯本）扔下布鲁克林大桥。小蜘蛛发射蛛丝一下子拉住她，下坠的冲力将她的颈椎骨硬生生扯断。格温从此与世长辞。电影最后格温穿了和漫画中死亡场景一模一样的衣服。
·谢琳·伍德蕾原本在片中扮演了玛丽·简的角色，并且还拍摄了一些镜头。但是在剪辑的时候，她的戏份全部被剪切掉了。原因是导演希望蜘蛛侠和格温·斯黛西之间的爱情能保留到最后。
·马克·韦伯在福克斯探照灯影业还有片约未完成。探照灯同意马克·韦伯执导索尼的《蜘蛛侠》，但条件就是要帮《X战警》宣传一下，所以就有了今年《X战警》的这个彩蛋。国内版无该片尾彩蛋。
·在电梯前支走Max去修电路的那名Oscorp员工，Max管他叫“斯迈思”（Smythe）。漫画中Smythe是“蜘蛛杀手”的创造者，后来他改造自己的身体，成为了“终极蜘蛛杀手”。
·Oscorp那名低调的秘书Felicia就是漫画中的黑猫，一个魅惑的亦正亦邪的角色，Peter在作为蜘蛛侠时的暧昧对象。
·Oscorp的地下仓库除了小绿魔和犀牛人的盔甲还有秃鹫和章鱼博士的装备。章鱼博士、秃鹫、电光人、猎人克拉文、致幻客与沙人在漫画中合称“Sinister Six”（邪恶六人组）。
·Peter的手机铃声是老版蜘蛛侠第一部中在地铁站扛着吉他卖艺的那哥们儿唱的歌“Spiderman”。 此旋律最初出处为1967年动画版蜘蛛侠的主题曲。
·《超凡蜘蛛侠》是一部用数码拍摄的影片，但导演选择了使用35mm胶片拍摄续集《超凡蜘蛛侠2》。而在拍摄结束之后，他会使用数字技术将胶片转换成数码影像，并用后期转制的方法把影片变成3D格式。
